Development has progressed through early- and mid-stage human studies in adults with obesity and with type 2 diabetes. Published phase 2 data reported reductions in body weight and improvements in glycemic markers over the treatment period. No regulatory agency has approved the compound for any indication, and it remains available only within controlled research settings. Whether benefits observed in trials translate into durable outcomes after treatment stops is not yet established.
Retatrutide is an investigational synthetic peptide that acts as an agonist at three distinct G protein-coupled receptors. It combines activity at the glucagon-like peptide-1 receptor, the glucose-dependent insulinotropic polypeptide receptor, and the glucagon receptor within a single molecule. This multi-receptor profile distinguishes it from earlier incretin-based compounds that engage one or two of these pathways. Researchers designed the molecule to test whether simultaneous activation produces greater metabolic effects than single or dual agonism alone.
The peptide backbone is chemically modified to resist rapid enzymatic breakdown in the body. A fatty acid side chain promotes binding to serum albumin, which slows renal clearance and supports an extended circulation time. These modifications allow less frequent administration than would be possible with an unmodified peptide. The precise contribution of glucagon receptor activation to the overall metabolic effect remains an area of active investigation, because glucagon raises glucose while also increasing energy expenditure.
Identity and purity are established with reversed-phase high-performance liquid chromatography and mass spectrometry. Chromatographic profiles reveal related impurities, truncated sequences, and oxidation products, while mass measurement confirms the expected molecular mass. Purity values for research material are typically reported as a percentage by peak area. Reference standards help calibrate retention behavior across instruments. Independent laboratories emphasize method suitability because results depend heavily on column chemistry, gradient, and detection wavelength. Batch-to-batch comparison relies on the same validated method.
Investigational peptide material is commonly distributed as a lyophilized powder in sealed vials. The solid form appears as a white to off-white cake or powder and is hygroscopic once opened. Peptides of this size are sensitive to moisture, repeated freeze-thaw cycles, and prolonged exposure to ambient light. Handling practices therefore emphasize desiccation, minimal vial opening, and cold storage. Working aliquots are often prepared to avoid repeatedly warming the bulk container.
| Property | Value | Notes |
|---|---|---|
| Molecular class | Synthetic peptide | Not a small molecule |
| Receptor targets | GLP-1, GIP, glucagon | Triple agonist activity |
| Appearance | White to off-white powder | Lyophilized solid |
| Solubility | Soluble in water | Also in aqueous buffers |
| Storage | −20 °C or below | Protect from light and moisture |
Peptides in this class degrade mainly through hydrolysis, oxidation, and aggregation. The lipid modification improves plasma residence time but can also promote self-association in aqueous solution at higher concentrations. Oxidation of methionine and deamidation of asparagine residues are common chemical liabilities that accumulate during storage. Stability studies therefore track purity loss, aggregate formation, and changes in receptor-binding potency over time under defined temperature and humidity conditions.
Solid peptide is generally held as a lyophilised powder at low temperature to slow degradation, with desiccant to limit moisture uptake. Reconstituted solutions are less stable and are usually kept refrigerated and protected from light for short periods. Repeated freeze-thaw cycles are avoided because they encourage aggregation. Laboratory handling includes work in a fume hood or laminar flow cabinet to limit inhalation and contamination. Weighing and transfer steps are performed with antistatic tools to reduce static-driven loss of fine powder.
Characterising a peptide of this size relies on a combination of chromatographic and mass spectrometric techniques. Reversed-phase high-performance liquid chromatography separates the intact molecule from related impurities, while electrospray mass spectrometry confirms molecular mass and detects truncation or oxidation products. Peptide mapping after enzymatic digestion verifies the amino acid sequence and locates modified residues. Because the molecule carries a lipid chain, assays must also distinguish the correctly conjugated product from incompletely acylated species.
Identity and purity are established by instrumental methods rather than by appearance. Reversed-phase high-performance liquid chromatography separates the peptide from related impurities and yields a purity value, usually expressed as the share of total peak area. Mass spectrometry checks that the observed mass agrees with the mass calculated from the published sequence, while peptide mapping or amino acid analysis adds structural evidence. Water content, counter-ion identity and residual solvents are sometimes reported as well. A certificate of analysis should name the method behind each figure, because results are method-dependent.
Laboratory handling follows the conventions used for other synthetic peptides. Lyophilized material is weighed and dissolved in an aqueous diluent, typically sterile water or bacteriostatic water, using gentle swirling rather than vigorous shaking, because foaming stresses the chain. Solutions are prepared under clean conditions and, where sterility matters, passed through a suitable filter. Working portions are kept small so that stock material is not repeatedly warmed and cooled, a practice that limits both aggregation and gradual loss of activity.
Alton Meister (1922–1995) was an American biochemist who made pioneering contributions to the study of glutathione metabolism. Alton Meister was born in New York City to Morris Meister and Florence Glickstein Meister. He received an undergraduate degree from Harvard University and an MD from Cornell University Medical College (now Weill Cornell Medical College). He then moved to the National Cancer Institute at the National Institutes of Health in Bethesda, Maryland. He remained there until 1955 when he became Chairman of the Department of Biochemistry at Tufts University. Meister returned to Cornell University Medical College in 1967 and served as chairman of its biochemistry department until 1991. He died in 1995 at the age of 72.
In the late 1920s, Pauling began publishing papers on the nature of the chemical bond. Between 1937 and 1938, he took a position as George Fischer Baker Non-Resident Lecturer in Chemistry at Cornell University. While at Cornell, he delivered a series of nineteen lectures and completed the bulk of his famous textbook The Nature of the Chemical Bond. It is based primarily on his work in this area that he received the Nobel Prize in Chemistry in 1954 "for his research into the nature of the chemical bond and its application to the elucidation of the structure of complex substances". Pauling's book has been considered "chemistry's most influential book of this century and its effective bible". In the 30 years after its first edition was published in 1939, the book was cited more than 16,000 times. Even today, many modern scientific papers and articles in important journals cite this work, more than seventy years after the first publication. Part of Pauling's work on the nature of the chemical bond led to his introduction of the concept of orbital hybridization. While it is normal to think of the electrons in an atom as being described by orbitals of types such as s and p, it turns out that in describing the bonding in molecules, it is better to construct functions that partake of some of the properties of each.
=== Lifestyle interventions === Individuals with all stages of the CKM syndrome have reduced chances of entering or progressing to stages 1 to 4 using lifestyle interventions. These interventions include eating a healthy diet, health-promoting physical activity (e.g., doing at leaset 150 minutes/week of aerobic exercise plus 2-3 strength trainings), and dietary-based weight loose including bariatric surgery if dietary methods do not reduce bodily weight.
== Genetics == The biologic instructions for synthesising protein C in humans are encoded in the gene officially named "protein C (inactivator of coagulation factors Va and VIIIa)". The gene's symbol approved by the HUGO Gene Nomenclature Committee is "PROC" from "protein C". It is located on the second chromosome (2q13-q14) and comprises nine exons. The nucleotide sequence that codes for human protein C is approximately 11,000 bases long.
Polymers for which UCST relies on non-ionic interactions are very sensitive to ionic contamination. Small amounts of ionic groups may suppress phase separation in pure water. The UCST is dependent on the molecular mass of the polymer. For the LCST this is not necessarily the case, as shown for poly(N-isopropylacrylamide).

=== Trump administration === On the same day of Powell's announcement, Trump denied having any knowledge of the investigation; White House Press Secretary Karoline Leavitt asserted that Trump did not order the Justice Department to investigate Powell. After suggesting at press conferences and social media posting in August and December 2025 that the administration would sue Powell due to the cost overruns, and after Powell announced that he was being targeted with an investigation, Trump said that Powell "either is incompetent or ... crooked" because of the cost overruns, and said that Powell will "be gone soon." However, Trump also said the investigation would not affect the timing of appointing a new Federal Reserve Chair as a successor to Powell. In an interview with Reuters on January 14, Trump said that he has no plans to fire Powell and that it was "too early" to say whether he would have legal grounds to remove Powell. Trump also dismissed concerns that diminished Federal Reserve independence would lead to higher inflation, asserting that "A president should have something to say" about monetary policy, while criticizing Republican Senators who may oppose his future Federal Reserve nominations due to the investigation, calling them disloyal. Secretary of the Treasury Scott Bessent has reportedly expressed concern that the investigation will cause volatility in the markets. While unnamed sources within the White House have suggested Federal Housing Finance Agency Director Bill Pulte may be behind the investigation of Powell, Pulte has publicly denied the allegations.
==== Antisense oligomer delivery ==== Antisense oligonucleotides (asONs) have been used in basic research and are being developed as possible medical treatments. CPP strategies have been developed to deliver antisense oligomers such as PNA and PMO into cells. Overcoming the repulsion by the cell membrane of negative-charged ONs and the degradation of asONs by enzymes, CPPs increase asONs bioavailability. Two types of neutral ON analogues, peptide nucleic acid (PNA) and phosphorodiamidate morpholino oligomers (PMO or Morpholino) are becoming dominant in this area. PNA has been conjugated with various CPPs either through disulfide linkages or through stable amide bonds. For example, antisense activity within cells that blocked expression of the galanin receptor was observed when a 21-mer PNA was coupled to the penetratin. Results on antiviral activity with PNA targeting HIV-1 have also been reported through disulfide linkage with TAT. CPP-PMO conjugates have also been successfully used to inhibit the replication of several viruses such as SARS and influenza and attachment of CPPs has improved the efficacy of splice-modifying Morpholinos in development for treatment of Duchenne muscular dystrophy
Salvinorin A is one of several structurally related salvinorins found in the Salvia divinorum plant. Salvinorin A is the only naturally occurring salvinorin that is known to be psychoactive. Salvinorin A can be synthesized from salvinorin B by acetylation, and de-acetylated salvinorin A becomes analog to salvinorin B. Research has produced a number of semi-synthetic compounds. Most derivatives are selective kappa opioid agonists as with salvinorin A, although some are even more potent, with the most potent compound salvinorin B ethoxymethyl ether being ten times stronger than salvinorin A. Some derivatives, such as herkinorin, reduce kappa opioid action and instead act as mu opioid agonists. The synthetic derivative RB-64 is notable because of its functional selectivity and potency. Salvinorin B methoxymethyl ether is seven times more potent than salvinorin A at KOPr in GTP-γS assays.
=== LS-SCLC === Chest X-rays are typically the first step to evaluate someone for any type of lung cancer. If images show suspicious spots on the patient's lung, a healthcare provider may order chest CT, PET, needle biopsy or bronchoscopy for further check. It is possible to use bronchoscopic biopsy to diagnose Lung small-cell carcinoma. However, small-cell carcinoma tissue obtained through bronchoscopy is prone to tissue compression and unclear morphology. However, pathologists can stain lesions with immunohistochemistry Ki-67, CD56, TTF-1, CgA, Syn, P63, CK5/6, LCA, and 34βE12 to help, in order to make a differential diagnosis.

Retatrutide is an investigational synthetic peptide that activates three metabolic receptors: GLP-1, GIP, and glucagon. It is being studied for obesity and type 2 diabetes. It has not been approved for clinical use.
Earlier incretin therapies engage one or two receptors. Retatrutide adds glucagon receptor activity, which may raise energy expenditure. Whether this produces larger clinical benefits is still being tested.
It has completed phase 2 trials and moved into later-stage evaluation. No regulator has granted approval. Access outside research settings is not authorized.
Solid powder is held frozen at -20 °C or below in a desiccated container. Reconstituted solutions are refrigerated and used within a limited period.
