The short version of triple agonist fits in a sentence. The long version — which is the one that helps — is below.
Reviewed 2026-04-18. Anything still debated is marked as such rather than presented as settled.
Clinical development has progressed through phase 2 trials in adults with obesity and type 2 diabetes, with phase 3 programs reported as ongoing. Reported outcomes include reductions in body weight and improvements in glycemic measures over defined treatment periods. Whether these effects translate into durable benefits after treatment ends remains an open question. Long-term safety data across broad populations are not yet complete, and regulatory decisions have not been announced.
Retatrutide is an investigational synthetic peptide engineered to activate three distinct hormone receptors within a single molecule. It targets the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor simultaneously. This triagonist design distinguishes it from earlier incretin-based compounds that act on one or two of these pathways. Structural modifications relative to native gut hormones extend its residence time in circulation. The molecule remains under clinical evaluation and is not approved for any indication.
Development has progressed from single- and multiple-ascending-dose studies in healthy volunteers into larger randomized trials in adults with obesity, type 2 diabetes, and fatty liver disease. Early reports describe dose-dependent reductions in body weight and improvements in glycemic markers over treatment periods of several months. Whether the glucagon arm adds tolerability cost without added benefit is still debated. Long-term cardiovascular outcomes, effects after treatment stops, and performance in older or comorbid populations are open questions rather than settled findings. Approval status may change, so the current investigational label should be confirmed against regulatory sources.
Retatrutide is an investigational synthetic peptide developed under the code LY3437943, with a backbone derived from glucose-dependent insulinotropic polypeptide. Several non-proteinogenic residues, including alpha-aminoisobutyric acid, appear in that backbone, and a fatty diacid side chain attached through a linker extends circulation time. The molecule carries roughly thirty-nine amino acid units and a total mass near 4.7 kilodaltons. Administration is by subcutaneous injection once weekly. Published work uses both the name retatrutide and the code LY3437943.

The intended pharmacology combines three signals in one molecule. GLP-1 receptor activation reduces appetite and slows gastric emptying, effects already exploited by approved incretin-based therapies. GIP receptor engagement is associated with improved insulin sensitivity and with direct effects on adipose tissue, although how much it contributes to overall outcomes is still debated. Glucagon receptor agonism raises energy expenditure and supports hepatic lipid handling, a mechanism that also tends to increase glucose output. The triple profile is hypothesized to produce a larger metabolic effect than single or dual agonism, but the relative weight of each receptor in humans is not settled.
Human evidence remains limited to controlled studies. A phase 2 trial in adults with obesity reported large, dose-dependent reductions in body weight over 48 weeks, with gastrointestinal events as the most frequently recorded adverse effect. Phase 3 programs designated TRIUMPH, for obesity, and TRANSCEND, for type 2 diabetes, are intended to confirm efficacy and to characterize safety in larger populations. Related studies are examining conditions such as knee osteoarthritis in people with obesity and metabolic liver disease. Open questions include long-term tolerability, effects on lean mass, and what happens after treatment is stopped.
Retatrutide is an investigational synthetic peptide that acts on three receptor targets at once: glucose-dependent insulinotropic polypeptide, glucagon-like peptide-1, and glucagon. It is developed by Eli Lilly and appears in the literature and in trial registries under the code LY3437943. The molecule belongs to a class of engineered peptides designed to resist rapid breakdown and permit infrequent subcutaneous administration. No regulatory agency has approved it for clinical use, and all available human data come from controlled trials rather than from routine practice.
Each receptor contributes a different physiological effect. Activation of the GLP-1 receptor slows gastric emptying and reduces appetite signaling in the brain. GIP receptor activity influences insulin secretion and lipid handling, while glucagon receptor stimulation raises energy use and fat oxidation. Combining these pathways is intended to produce weight loss beyond what single- or dual-receptor agonists achieve. Researchers attribute the observed potency to simultaneous engagement of all three targets, though the exact contribution of each receptor to overall effect remains under investigation.
Clinical development has advanced through phase 2 trials in adults with obesity and type 2 diabetes. Reported phase 2 results described substantial average weight reduction over roughly forty-eight weeks of weekly dosing. A phase 3 program is ongoing to confirm efficacy and assess long-term safety. Because the compound has not received regulatory approval, it is not available as a prescription product. Public discussion of retatrutide often conflates trial findings with marketed status, an important distinction when interpreting coverage of the topic.

Fermium was first discovered in the fallout from the 'Ivy Mike' nuclear test (1 November 1952), the first successful test of a hydrogen bomb. Initial examination of the debris from the explosion had shown the production of a new isotope of plutonium, 244Pu: this could only have formed by the absorption of six neutrons by a uranium-238 nucleus followed by two β− decays. At the time, the absorption of neutrons by a heavy nucleus was thought to be a rare process, but the identification of 244Pu raised the possibility that still more neutrons could have been absorbed by the uranium nuclei, leading to new elements. Element 99 (einsteinium) was quickly discovered on filter papers which had been flown through clouds from the explosion (the same sampling technique that had been used to discover 244Pu). It was then identified in December 1952 by Albert Ghiorso and co-workers at the University of California at Berkeley. They discovered the isotope 253Es (half-life 20.5 d) that was made by the capture of 15 neutrons by uranium-238 nuclei – which then underwent seven successive beta decays:
Traditional food cultures reflect local variations in tolerance and historically many societies have adapted to low levels of tolerance by making dairy products that contain less lactose than fresh milk. One ethnographic example of this is kumis, a fermented milk product that contains little to no lactose, which is the main source of dairy nutrition in Mongolia. The medicalization of lactose intolerance as a disorder has been attributed to biases in research history, since most early studies were conducted amongst populations which are normally tolerant, as well as the cultural and economic importance and impact of milk in countries such as the United States.
== Production and properties == Butyl acrylate can be produced by the acid-catalyzed esterification of acrylic acid with butanol. It polymerizes easily, therefore commercial preparations contain polymerization inhibitors such as hydroquinone, phenothiazine, or hydroquinone ethyl ether.
A worldwide summary of more than one hundred human studies reported a median of 22.1 μmol/L for serum α-tocopherol and defined α-tocopherol deficiency as less than 12 μmol/L. It cited a recommendation that serum α-tocopherol concentration be ≥30 μmol/L to optimize health benefits. In contrast, the U.S. Dietary Reference Intake text for vitamin E concluded that a plasma concentration of 12 μmol/L was sufficient to achieve normal ex vivo hydrogen peroxide-induced hemolysis. A 2014 review defined less than 9 μmol/L as deficient, 9-12 μmol/L as marginal, and greater than 12 μmol/L as adequate. Regardless of which definition is used, vitamin E deficiency is rare in humans, occurring as a consequence of abnormalities in dietary fat absorption or metabolism rather than from a diet low in vitamin E. Cystic fibrosis and other fat malabsorption conditions can result in low serum vitamin E. One example of a genetic abnormality in metabolism is mutations of genes coding for alpha-tocopherol transfer protein (α-TTP). Humans with this genetic defect exhibit a progressive neurodegenerative disorder known as ataxia with vitamin E deficiency (AVED) despite consuming normal amounts of vitamin E. Large amounts of alpha-tocopherol as a dietary supplement are needed to compensate for the lack of α-TTP. Bariatric surgery as a treatment for obesity can lead to vitamin deficiencies. Long-term follow-up reported a 16.5% prevalence of vitamin E deficiency. There are guidelines for multivitamin supplementation, but adherence rates are reported to be less than 20%.

The female lays three to seven eggs, which both sexes help to incubate. Its breeding range extends from central and western Alaska across northern Canada to Nova Scotia, and south throughout North America, into central Mexico and the Caribbean. It is a local breeder in Central America and is widely distributed throughout South America. Most birds breeding in Canada and the northern United States migrate south in the winter. It is an occasional vagrant to Western Europe. The American kestrel is often used in falconry, especially by beginners. Though not as strong a flyer as many other, larger falcons, proper training and weight control by the falconer can allow captive American kestrels to become effective hunters of birds in the size range of sparrows and starlings, with occasional success against birds up to approximately twice their own weight.
== Other media == A Borderlands film was released by Gearbox and Lionsgate in 2024, with Eli Roth as director. The film received negative reviews from critics and poor reception from fans. In April 2020, Gearbox announced it was developing a television series based on its Brothers in Arms series.
Ethylene oxide is an organic compound with the formula C2H4O. It is a cyclic ether and the simplest epoxide: a three-membered ring consisting of one oxygen atom and two carbon atoms. Ethylene oxide is a colorless and flammable gas with a faintly sweet odor. Because it is a strained ring, ethylene oxide easily participates in a number of addition reactions that result in ring-opening. Ethylene oxide is isomeric with acetaldehyde and with vinyl alcohol. Ethylene oxide is industrially produced by oxidation of ethylene in the presence of a silver catalyst. The reactivity that is responsible for many of ethylene oxide's hazards also makes it useful. Although too dangerous for direct household use and generally unfamiliar to consumers, ethylene oxide is used for making many consumer products as well as non-consumer chemicals and intermediates. These products include detergents, thickeners, solvents, plastics, and various organic chemicals such as ethylene glycol, ethanolamines, simple and complex glycols, polyglycol ethers, and other compounds. Although it is a vital raw material with diverse applications, including the manufacture of products like polysorbate 20 and polyethylene glycol (PEG) that are often more effective and less toxic than alternative materials, ethylene oxide itself is a very hazardous substance. At room temperature it is a very flammable, carcinogenic, mutagenic, irritating; and anaesthetic gas.

== Interactions == Trimipramine should not be given with sympathomimetic agents such as epinephrine (adrenaline), ephedrine, isoprenaline, norepinephrine (noradrenaline), phenylephrine and phenylpropanolamine. Barbiturates may increase the rate of metabolism. Trimipramine should be administered with care in patients receiving therapy for hyperthyrodism.
=== Vaccines === The cold chain is used in the supply of vaccines to distant clinics in hot climates served by poorly developed transport networks. Disruption of a cold chain due to war or logistical challenges may produce severe consequences, as distributed vaccines can become inert due to a lack of temperature control during transport. The integrity of the vaccine cold chain is therefore a critical public health concern. For vaccines, there are different types of cold chains. There is an ultralow, or deep freeze, cold chain for vaccines that require -70 degrees C, such as the Ebola and Pfizer–BioNTech COVID-19 vaccines, and some animal vaccines. Next the frozen chain requires -20 degrees C. Varicella and zoster vaccinations require this level. Then the refrigerated chain, which requires temperatures between two and eight degrees C. Most flu vaccinations only require refrigeration. In 2020, during the COVID-19 pandemic, vaccines being developed may need ultracold storage and transportation temperatures as cold as −70 °C (−94 °F), requiring what has been referred to as a "colder chain" infrastructure. This creates some issues of distribution for the Pfizer vaccine. It is estimated that only 25 to 30 countries in the world have the infrastructure for the required ultracold cold chain.
In the late 19th century, a style known as anthropomorphic taxidermy became popular. A 'Victorian whimsy', mounted animals were dressed as people or displayed as if engaged in human activities. An early example of this genre was displayed by Herman Ploucquet, from Stuttgart, Germany, at the Great Exhibition in London.
=== Noncovalent gas phase interactions === Electrospray ionization is also utilized in studying noncovalent gas phase interactions. The electrospray process is thought to be capable of transferring liquid-phase noncovalent complexes into the gas phase without disrupting the noncovalent interaction. Problems such as non specific interactions have been identified when studying ligand substrate complexes by ESI-MS or nanoESI-MS. An interesting example of this is studying the interactions between enzymes and drugs which are inhibitors of the enzyme. Competition studies between STAT6 and inhibitors have used ESI as a way to screen for potential new drug candidates. Electrospray ionization can even be used for studying protein complexes >1 MDa.
A variety of circumstances and drugs have been found to affect oxytocin levels in humans. Labor, breastfeeding, pregnancy, hypoglycemia, sexual self-stimulation, physical activity, and psychosocial stress all increase oxytocin levels in humans. Oxytocin levels increase 3- to 4-fold during pregnancy and increase a further 3- to 4-fold during labor. They also increase by 5-fold during breastfeeding. In premenopausal women, oxytocin levels are almost twice as high during ovulation and the luteal phase than during the follicular phase. Oxytocin levels are higher, by about 2-fold, in new romantic relationships. Levels of oxytocin are lower in underweight women with anorexia nervosa, returning to normal with recovery. Estrogens such as estradiol and ethinylestradiol have been found to dose-dependently and robustly increase oxytocin levels in humans. In addition, they have been found to increase the expression of the oxytocin receptor in the brain. Controlled ovarian hyperstimulation can increase estradiol levels by around 90-fold and oxytocin levels by about 6.5-fold. Increased estrogen levels are thought to be involved in the increases in oxytocin levels during the ovulatory and luteal phases, during pregnancy, and in the premenopause compared to the postmenopause. Whereas estrogens unambiguously stimulate oxytocin production, findings appear to be mixed on the influence of androgens such as testosterone on oxytocin production and oxytocin receptor expression.

It is a synthetic peptide triagonist that engages GIP, GLP-1, and glucagon receptors. Investigational compounds in this class are studied for metabolic conditions rather than for a single organ system.
No regulatory agency has approved it for any indication. It remains an investigational product studied within controlled clinical trial programs.
A dual agonist engages two receptor targets, typically GIP and GLP-1. A triagonist adds glucagon receptor activity, which is intended to influence energy expenditure alongside appetite and glucose handling.
As of the mid-2020s retatrutide remains investigational and is not an approved medicine in the United States or the European Union. It has been supplied mainly to participants in clinical trials. Labels and availability can change, so regulatory listings should be checked directly.
