The short version of peptide backbone fits in a sentence. The long version — which is the one that helps — is below.

Several questions remain unresolved. It is not yet known whether the compound reduces cardiovascular events or mortality, because outcome studies require long follow-up. The durability of weight reduction after treatment withdrawal is uncertain, and rebound has been observed with other incretin-based therapies. Long-term safety data covering several years are limited. Effects in adolescents, in pregnancy, and in people with significant kidney or liver impairment have not been characterized in published reports.
Randomized studies of retatrutide measure change in body weight as a percentage of baseline, along with absolute weight loss. Glycemic endpoints include hemoglobin A1c and fasting plasma glucose. Investigators also track blood pressure, lipid fractions, and liver fat content to characterize effects beyond weight alone. Trial designs typically use double-blind, placebo-controlled groups with periodic dose escalation, and they record adverse events throughout both treatment and follow-up periods.
Development has progressed through early- and mid-stage human studies in adults with obesity and with type 2 diabetes. Published phase 2 data reported reductions in body weight and improvements in glycemic markers over the treatment period. No regulatory agency has approved the compound for any indication, and it remains available only within controlled research settings. Whether benefits observed in trials translate into durable outcomes after treatment stops is not yet established.
Retatrutide is an investigational synthetic peptide that acts as an agonist at three distinct G protein-coupled receptors. It combines activity at the glucagon-like peptide-1 receptor, the glucose-dependent insulinotropic polypeptide receptor, and the glucagon receptor within a single molecule. This multi-receptor profile distinguishes it from earlier incretin-based compounds that engage one or two of these pathways. Researchers designed the molecule to test whether simultaneous activation produces greater metabolic effects than single or dual agonism alone.
The peptide backbone is chemically modified to resist rapid enzymatic breakdown in the body. A fatty acid side chain promotes binding to serum albumin, which slows renal clearance and supports an extended circulation time. These modifications allow less frequent administration than would be possible with an unmodified peptide. The precise contribution of glucagon receptor activation to the overall metabolic effect remains an area of active investigation, because glucagon raises glucose while also increasing energy expenditure.

Identity and purity are established by instrumental methods rather than by appearance. Reversed-phase high-performance liquid chromatography separates the peptide from related impurities and yields a purity value, usually expressed as the share of total peak area. Mass spectrometry checks that the observed mass agrees with the mass calculated from the published sequence, while peptide mapping or amino acid analysis adds structural evidence. Water content, counter-ion identity and residual solvents are sometimes reported as well. A certificate of analysis should name the method behind each figure, because results are method-dependent.
Laboratory handling follows the conventions used for other synthetic peptides. Lyophilized material is weighed and dissolved in an aqueous diluent, typically sterile water or bacteriostatic water, using gentle swirling rather than vigorous shaking, because foaming stresses the chain. Solutions are prepared under clean conditions and, where sterility matters, passed through a suitable filter. Working portions are kept small so that stock material is not repeatedly warmed and cooled, a practice that limits both aggregation and gradual loss of activity.
Stability depends strongly on physical state. Dry powder is comparatively robust when held at -20 °C or below, desiccated and shielded from light; under those conditions degradation is slow and measured over years. Once dissolved, the peptide becomes far more vulnerable: backbone hydrolysis, oxidation of susceptible residues and aggregation all proceed faster in solution, and the rates climb with temperature and with pH far from neutral. Refrigerated storage at 2–8 °C extends usable life for short periods, and repeated freeze–thaw cycles are best avoided.
Human evidence remains limited to controlled studies. A phase 2 trial in adults with obesity reported large, dose-dependent reductions in body weight over 48 weeks, with gastrointestinal events as the most frequently recorded adverse effect. Phase 3 programs designated TRIUMPH, for obesity, and TRANSCEND, for type 2 diabetes, are intended to confirm efficacy and to characterize safety in larger populations. Related studies are examining conditions such as knee osteoarthritis in people with obesity and metabolic liver disease. Open questions include long-term tolerability, effects on lean mass, and what happens after treatment is stopped.
Retatrutide is an investigational synthetic peptide that acts on three receptor targets at once: glucose-dependent insulinotropic polypeptide, glucagon-like peptide-1, and glucagon. It is developed by Eli Lilly and appears in the literature and in trial registries under the code LY3437943. The molecule belongs to a class of engineered peptides designed to resist rapid breakdown and permit infrequent subcutaneous administration. No regulatory agency has approved it for clinical use, and all available human data come from controlled trials rather than from routine practice.
The intended pharmacology combines three signals in one molecule. GLP-1 receptor activation reduces appetite and slows gastric emptying, effects already exploited by approved incretin-based therapies. GIP receptor engagement is associated with improved insulin sensitivity and with direct effects on adipose tissue, although how much it contributes to overall outcomes is still debated. Glucagon receptor agonism raises energy expenditure and supports hepatic lipid handling, a mechanism that also tends to increase glucose output. The triple profile is hypothesized to produce a larger metabolic effect than single or dual agonism, but the relative weight of each receptor in humans is not settled.
Quality control of research material relies on several complementary checks. Purity testing confirms the absence of truncated or oxidized peptide species, while water content and counterion analysis show how much mass comes from salts rather than the peptide itself. Sequence verification through tandem mass spectrometry ensures the correct amino acid chain. Because unregulated suppliers vary widely, independent verification of identity and purity is often necessary before a sample enters experiments.
Documentation plays a practical role in maintaining consistent results across laboratories. Certificates of analysis list purity, identity, and testing methods, and batch numbers allow comparisons between lots. Records of storage temperature and handling history help investigators interpret unexpected findings. When a sample behaves anomalously, reviewing that documentation often reveals whether the cause lies in the material or in the assay conditions.
Laboratories identify and quantify retatrutide using reversed-phase high-performance liquid chromatography coupled to mass spectrometry. This approach separates the peptide from related impurities and confirms identity through mass-to-charge measurements. Purity is commonly reported as the area percentage of the main peak relative to the total chromatogram. Ultraviolet detection near 214 nanometers is also used for peptide quantification, while intact mass analysis checks the molecular weight against a reference value.
Commonly used salts in lysing buffers include: a. Sodium chloride (NaCl): NaCl is often included to maintain isotonic conditions, preventing osmotic shock and cell rupture during the lysis process. b. Potassium chloride (KCl): Similar to NaCl, KCl can be used to adjust the ionic strength and facilitate cell lysis. Enzymes: Certain enzymes are added to lysing buffers to enhance cell lysis by digesting specific cellular components that can interfere with the extraction of the target enzyme. Examples of enzymes used in lysing buffers include: a. Lysozyme: Lysozyme breaks down the peptidoglycan layer of bacterial cell walls, weakening their structural integrity and facilitating subsequent disruption. It is particularly effective for Gram-positive bacteria. b. DNase (Deoxyribonuclease): DNase degrades DNA present in the lysate, reducing its viscosity and preventing DNA-related interference in downstream purification steps. c. RNase (Ribonuclease): Similar to DNase, RNase degrades RNA in the lysate, reducing its viscosity and minimizing RNA-related interference. The specific combination and concentrations of detergents, salts, and enzymes in lysing buffers can vary depending on the target enzyme, cell type, and experimental requirements, optimization of these components is crucial to achieve efficient cell lysis while preserving the stability and activity of the desired enzyme during the purification process.
==== Earliest research ==== Charles P. Slichter wrote that "In 1955, Redfield showed that the conventional theory of saturation did not properly account for the experimental facts of nuclear resonance in solids...[Redfield] showed that the conventional approach essentially defied the second law of thermodynamics." Redfield studied NMR with Charles Pence Slichter, assisting with early superconductivity experiments at University of Illinois, Urbana and published the Redfield Theory as a postdoc under Nicolaas Bloembergen at Harvard. At first he studied electron removal in argon, hydrogen and crypton, and the movement of electrons in photoconductors, including his doctoral thesis on the Hall effect in diamonds and salt crystals. After his breakthrough work on relaxation theory, he continued to produce papers on nuclear spin relaxation.
== Political positions == As a party in Albania, the Socialist Party of Albania is described as social-democratic and centre-left. In its 2013 party platform, the party has pledged to replace the flat tax with a progressive tax, and also supports universal health care. Party leader Edi Rama has indicated that he supports LGBT rights in Albania. The party is also pro-European, supports the accession of Albania to the European Union, NATO membership, and considers Kosovo "Albania's main strategic partner and ally".
== Toxicity and deficiency == There is an optimal range of selenium concentration in animals (or broadly speaking, in almost every selenium-dependent lifeform). Too much results in toxicity, too little results in deficiency.
Tralokinumab sold under the brand names Adtralza (EU/UK) and Adbry (US) among others, is a human monoclonal antibody used for the treatment of atopic dermatitis. The broadcast pattern covered an area of about 100 degrees in a roughly fan-shaped area, with a smaller side lobe to the rear, courtesy of the reflectors, and much smaller ones to the sides. When the signal reflected off the ground it underwent a ½ wavelength phase-change, which caused it to interfere with the direct signal. The result was a series of vertically stacked lobes about 5 degrees wide from 1 degree off the ground to the vertical. The system was later expanded by adding another set of four additional antennas closer to the ground, wired in a similar fashion. The receiver consisted of an Adcock array consisting of four 240 foot (73 m) tall wooden towers arranged at the corners of a square. Each tower had three sets (originally two) of receiver antennas, one at 45, 95 and 215 feet off the ground. The mean height of the transmitter stack was 215 feet, which is why the topmost antenna was positioned at the same altitude in order to produce a reception pattern that was identical to the transmission. A set of motor-driven mechanical switches allowed the operator to select which antenna was active. The output of the selected antenna on each of the four towers was sent to a single radiogoniometer system (not Watt's own huff-duff solution). By connecting the antennas together in X-Y pairs the horizontal bearing could be measured, while connecting together the upper and lower antennas allowed the same goniometer to be used to measure the vertical angle. Two physical layout plans were used, either 'East Coast' or 'West Coast'.
German philosophy is historically significant: Gottfried Leibniz's contributions to rationalism; the enlightenment philosophy by Immanuel Kant; the establishment of classical German idealism by Johann Gottlieb Fichte, Georg Wilhelm Friedrich Hegel and Friedrich Wilhelm Joseph Schelling; Arthur Schopenhauer's composition of metaphysical pessimism; the formulation of communist theory by Karl Marx and Friedrich Engels; Friedrich Nietzsche's development of perspectivism; Gottlob Frege's contributions to the dawn of analytic philosophy; Martin Heidegger's works on Being; Oswald Spengler's historical philosophy; and the development of the Frankfurt School have all been very influential.
=== Treatment and telemedicine === Providing treatment including prescribing medications based on remote information gathering without a proper established relationship is not accepted as good practice with few exceptions. These exceptions include cross-coverage within a practice and certain public health urgent or emergent issues. The ethics of telemedicine including questions on its impact to diagnosis, doctor-patient relationship, and continuity of care have been raised. However, with appropriate use and specific guidelines, risks may be minimized and the benefits including increased access to care may be realized.
=== Enzymatic method for RNA oligonucleotide synthesis === One approach under investigation for overcoming length limitations in oligonucleotide synthesis involves the use of engineered enzymes to construct RNA sequences, rather than relying solely on traditional chemical methods. A key enzyme studied in this context is CID1 poly(U) polymerase (PUP), which naturally catalyzes the addition of nucleotides to the 3′ end of an RNA strand. In its native form, PUP functions as a template-independent polymerase, typically adding repeated sequence nucleotide tails to RNA molecules. To enable controlled sequence synthesis, enzymatic RNA synthesis has been adapted into a cyclic two-step process consisting of nucleotide extension and deblocking. During the extension step, a single nucleotide is enzymatically added, while the deblocking step removes a protecting group to allow subsequent incorporation. One investigated method involves the use of nucleoside triphosphates modified with a 3′-O-allyl blocking group. These modified nucleotides are synthesized and purified using techniques such as ion-exchange chromatography and preparative high-performance liquid chromatography (HPLC). Upon incorporation by PUP, the 3′-O-allyl group prevents further extension, thereby enabling single-nucleotide addition per cycle. Chemical removal of the blocking group then permits the next round of extension. Studies have shown that wild-type PUP exhibits limited efficiency when incorporating modified nucleotides.
=== Evolution === The delicate skeletons of bats do not fossilise well; it is estimated that only 12% of bat genera that lived have been found in the fossil record. The oldest known bat fossils include Archaeonycteris praecursor and Altaynycteris aurora (55–56 million years ago), both known only from isolated teeth. The oldest complete bat skeletons are Icaronycteris gunnelli and Onychonycteris finneyi (52 million years ago), known from two skeletons discovered in Wyoming. The extinct bats Palaeochiropteryx and Hassianycteris from the Messel Pit of Germany, both of which lived 48 million years ago, are the first fossil mammals whose colouration has been discovered: both were reddish-brown. The largest known fossil bat known from a near-complete skeleton is Archaeopteropus (~33 million years ago) from Italy, which had a wingspan of about 82 to 90 centimetres (2.69 to 2.95 ft). Bats were formerly grouped in the superorder Archonta, along with the treeshrews (Scandentia), colugos (Dermoptera), and primates. Modern genetic evidence now places bats in the superorder Laurasiatheria, with its sister taxon as Ferungulata, which includes carnivorans, pangolins, odd-toed ungulates, and even-toed ungulates. One study places Chiroptera as a sister taxon to odd-toed ungulates (Perissodactyla).

=== Politics === During the 1990 campaign for governor of Florida, it was disclosed that one of the candidates, Lawton Chiles, had depression and had resumed taking fluoxetine, leading his political opponents to question his fitness to serve as governor. The 2005 Red Lake shootings were committed a week after the perpetrator's dosage of Prozac was allegedly increased. The murders and suicide renewed public debate about the safety of Prozac use among children and adolescents.
In the twenty-first century, donkey milk is largely used in the manufacture of soaps and moisturizers, but evidence shows it has possible medical uses to treat infants and children with cow's milk protein allergy (CMPA) as a natural "formula" for infants.
(...) To make matters worse, Admiral Thomas Cochrane (British), whose naval services and expenses had remained unpaid, appropriated the reserves of silver bars that had been painfully and arrogantly accumulated during the government of San Martín. Cochrane was the commander of the Chilean "liberation" fleet and also benefited from the capture and hijacking of Peruvian merchant ships. A French diplomat informed his bosses in Paris that the lack of popular support for freedom and independence was explained by the corruption of the new separatist authorities and their infighting. Another diplomatic envoy attributed the weakness of these nascent governments to the distribution of official positions through protection and intrigue instead of recognition of merit. These weak organizational bases provided fertile conditions for corruption and abuse of power." Later, during the founding of the State of Upper Peru, there were anti-Peruvian sectors in Argentina that saw the independence of Bolivia (and the renunciation of its claims by the United Provinces of the Río de la Plata to the sovereignty of that territory) as something tolerable with to avoid the aggrandizement of Peru (coinciding with Bolívar, Sucre and Santander to avoid restoring the power that Peru had during the viceregal era), which had been a great problem for the commercial and military interests of Buenos Aires during the wars that there was between the Junta and the Peruvian Viceroyalty.
== Treatment == The treatment aims to create an environment that allows skin to grow across an ulcer. In most cases, this requires finding and treating underlying venous reflux. The National Institute for Health and Care Excellence (NICE) recommends referral to a vascular service for anyone with a leg ulcer that has not healed within two weeks or anyone with a healed leg ulcer. Most venous ulcers respond to patient education, elevation of the foot, elastic compression, and evaluation (known as the Bisgaard regimen). Exercise together with compression stocking increases healing. There is no evidence that antibiotics, whether administered intravenously or by mouth, are useful. Silver products are also not typically useful, while there is some evidence of benefit from cadexomer iodine creams. There is a lack of quality evidence regarding the use of medical-grade honey for venous leg ulcers. The recommendations for dressings to treat venous ulcers vary between the countries. Antibiotics are often recommended to be used only if advised by the physician due to the emergence of bacterial resistance to antibiotics. This is an issue with venous ulcers as they tend to heal more slowly than acute wounds, for example. Natural alternatives that are suitable for long-term use exist on the market, such as honey and resin salve. These products are considered as Medical Devices in EU and the products have to be CE marked. Sugar has long been known for its effectiveness in wound treatment, notably through the use of honey or powdered sugar.
As the world's population continues to grow, experts believe that current food supplies will not be able to supply the population. Thus, a sustainable food source is critical. Studies have shown that entomophagy, the consumption of insects, has the potential to sustain a growing population. Insects such as crickets require less feed, less water, and provide around the same amount of protein that chickens, cows, and pigs do. Crickets can be ground into a protein flour. In one study, researchers provide an overview of the process of 3D printing insect flour into foods that do not resemble insects; thus, keeping the nutritional value of the insect intact.

Trials measure percentage change in body weight, absolute weight loss, and glycemic markers such as hemoglobin A1c. They also record blood pressure, lipids, and liver fat. Adverse events are tracked throughout.
Mass spectrometry identifies molecules by mass-to-charge transitions, which reduces interference from related peptides. Immunoassays run faster but can cross-react. Structural confirmation usually requires the mass spectrometric approach.
Cardiovascular outcomes, long-term safety, and weight regain after stopping treatment are unresolved. These questions need years of follow-up data. Published evidence covers only limited treatment durations.
