Retatrutide is an investigational synthetic peptide engineered to activate three distinct hormone receptors within a single molecule. It targets the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor simultaneously. This triagonist design distinguishes it from earlier incretin-based compounds that act on one or two of these pathways. Structural modifications relative to native gut hormones extend its residence time in circulation. The molecule remains under clinical evaluation and is not approved for any indication.
Receptor activation produces downstream effects that differ by tissue. GLP-1 receptor signaling influences appetite regulation and insulin secretion in a glucose-dependent manner. GIP receptor activity contributes to metabolic handling of nutrients and may modulate adipose tissue. Glucagon receptor engagement raises energy expenditure and promotes hepatic lipid turnover, though the balance among these actions in humans is still being characterized. Preclinical models showed reductions in body weight and improved glycemic markers.
Clinical development has progressed through phase 2 trials in adults with obesity and type 2 diabetes, with phase 3 programs reported as ongoing. Reported outcomes include reductions in body weight and improvements in glycemic measures over defined treatment periods. Whether these effects translate into durable benefits after treatment ends remains an open question. Long-term safety data across broad populations are not yet complete, and regulatory decisions have not been announced.
Body composition is assessed with dual-energy X-ray absorptiometry or comparable methods, which separate fat mass from lean mass. Reported losses include both compartments, and the ratio between them is a subject of ongoing analysis rather than a settled result. Waist circumference, blood pressure, and lipid panels are collected as supporting measures. Resting energy expenditure and substrate oxidation are measured in smaller mechanistic studies, where glucagon receptor activity is expected to matter. These substudies are typically short and small, so their findings carry wide uncertainty.
Interpretation depends on study phase and duration. Phase 2 programs are powered for weight and safety signals, not for cardiovascular or renal outcomes, which require event-driven designs. Gastrointestinal events such as nausea, diarrhea, vomiting, and constipation are the most frequently reported adverse effects and tend to cluster around dose escalation. Small increases in heart rate have been described. Because follow-up after treatment discontinuation is limited, questions about weight regain and durability are open rather than answered.

Published phase 2 data reported reductions in body weight and improvements in glycemic markers over the treatment period. No regulatory agency has approved the compound for any indication, and it remains available only within controlled research settings. Whether benefits observed in trials translate into durable outcomes after treatment stops is not yet established.
Retatrutide is an investigational synthetic peptide that acts as an agonist at three distinct G protein-coupled receptors. It combines activity at the glucagon-like peptide-1 receptor, the glucose-dependent insulinotropic polypeptide receptor, and the glucagon receptor within a single molecule. This multi-receptor profile distinguishes it from earlier incretin-based compounds that engage one or two of these pathways. Researchers designed the molecule to test whether simultaneous activation produces greater metabolic effects than single or dual agonism alone.
The peptide backbone is chemically modified to resist rapid enzymatic breakdown in the body. A fatty acid side chain promotes binding to serum albumin, which slows renal clearance and supports an extended circulation time. These modifications allow less frequent administration than would be possible with an unmodified peptide. The precise contribution of glucagon receptor activation to the overall metabolic effect remains an area of active investigation, because glucagon raises glucose while also increasing energy expenditure.
As a peptide, the compound is generally supplied as a lyophilized powder and stored frozen to slow degradation. Recommended conditions usually sit at minus twenty degrees Celsius or colder, shielded from light and moisture. Solutions are less stable than the dry powder and are often prepared fresh before analysis. Repeated freeze-thaw cycles can drive aggregation, so splitting stock material into small aliquots reduces handling stress and preserves sample integrity.
Quality control of research material relies on several complementary checks. Purity testing confirms the absence of truncated or oxidized peptide species, while water content and counterion analysis show how much mass comes from salts rather than the peptide itself. Sequence verification through tandem mass spectrometry ensures the correct amino acid chain. Because unregulated suppliers vary widely, independent verification of identity and purity is often necessary before a sample enters experiments.
Aldehyde tags were first inserted into the modified sulfatase motif peptide for proteins of interest in 2007. Since then, similar usage of aldehydes and ketones as chemical reporters in bioorthogonal applications has been demonstrated in self-assembly of cell-lysing drugs, the targeting of proteins, as well as glycans and the preparation of heterobifunctional fusion proteins.
=== Alcoholic drinks === In 2010, Starbucks began selling alcoholic beverages at some stores in the United States. In August 2014, Starbucks opened its first store in Williamsburg, Brooklyn which by then was among 30 locations serving beer and wine. In 2016, three locations within Toronto, Ontario also announced they were going to serve alcohol, including up-scale appetizers like bacon-wrapped dates and truffle mac and cheese. Starbucks ensures that the selected locations that serve alcohol are able to accommodate the community, as well as have the space for extra seating and storage.
Notes Compensation picks are selections in addition to the normal order of selection, allocated to clubs by the AFL as compensation for losing uncontracted players to the new expansion clubs, Gold Coast and Greater Western Sydney. The picks can be held for up to five years and clubs declare at the beginning of the season of their intent to utilise the pick at the end of the season. Picks could be traded to other clubs in return for players or other draft selections. Free agency compensation picks are additional selections awarded to teams based on their net loss of players during the free agency trade period. Academy players are local zone selections available to the four NSW and Queensland clubs. Both academy and father-son selections are subject to a bidding process, where the club with the family or academy connection must match any opposition club's bid with their next available selection.
The European Commission approved Inotuzumab ozogamicin as a monotherapy for the treatment of adults with relapsed or refractory CD22-positive B-cell precursor acute lymphoblastic leukemia (ALL) on June 30, 2017, under the brand name Besponsa (Pfizer/Wyeth), followed on August 17, 2017, by the FDA. The first immunology antibody–drug conjugate (iADC), ABBV-3373, showed an improvement in disease activity in a Phase 2a study of patients with rheumatoid arthritis and a study with the second iADC, ABBV-154 to evaluate adverse events and change in disease activity in participants treated with subcutaneous injection of ABBV-154 is ongoing. In July 2018, Daiichi Sankyo Company, Limited and Glycotope GmbH have inked a pact regarding the combination of Glycotope's investigational tumor-associated TA-MUC1 antibody gatipotuzumab and Daiichi Sankyo's proprietary ADC technology for developing gatipotuzumab antibody drug conjugate. In 2019 AstraZeneca agreed to pay up to US$6.9 billion to jointly develop DS-8201 with Japan's Daiichi Sankyo. It is intended to replace Herceptin for treating breast cancer. DS8201 carries eight payloads, compared to the usual four.

Poppy straw (also known as opium straw, mowed opium straw, crushed poppy capsule, poppy chaff, or poppy husk) is derived from opium poppies (Papaver somniferum) that are harvested when fully mature and dried by mechanical means. Opium poppy straw is what remains after the seed pods have been harvested — that is, the dried stalks, stem and leaves of poppies grown for their seeds. The field-dried leaves, stalk, and seed pod are then used in commercial manufacture of morphine or other poppy-alkaloid derived drugs, by first processing the material, separating the seeds, and then making concentrate of poppy straw where no extraction using the traditional methods of latex extraction has been made. The straw was originally considered an agricultural by-product of the mechanised poppy seed harvest, which was primarily grown for its edible and oil-producing seed. This changed in 1927 when János Kabay developed a chemical process to extract morphine from the crushed capsule. Concentrated poppy straw, consisting mainly of the crushed capsule without the seeds, soon became a valuable source of morphine. Today, concentrate of poppy straw is a major source of many opiates and other alkaloids. It is the source of 90% of the world supply of legal morphine (for medical and scientific use) and in some countries it also is a source of illegal morphine, which could be processed into illegal heroin. The 1961 Single Convention on Narcotic Drugs defines poppy straw as "all parts (except the seeds) of the opium poppy, after mowing".
Although as of 2016, NEO212 has not been tested in humans, it has shown superior cancer therapeutic activity in animal models of glioma, melanoma, and brain metastasis of triple-negative breast cancer. Because tumor cells that express the O-6-methylguanine-DNA methyltransferase (MGMT) gene are more resistant to the effects of temozolomide, researchers investigated whether the inclusion of O6-benzylguanine (O6-BG), an AGT inhibitor, could overcome this resistance and improve the drug's therapeutic effectiveness. In the laboratory, this combination indeed showed increased temozolomide activity in tumor-cell culture in vitro and in animal models in vivo. However, a recently completed phase-II clinical trial with brain-tumor patients yielded mixed outcomes; while there was some improved therapeutic activity when O6-BG and temozolomide were given to patients with temozolomide-resistant anaplastic glioma, there seemed to be no significant restoration of temozolomide sensitivity in patients with temozolomide-resistant glioblastoma multiforme. Some efforts focus on engineering hematopoietic stem cells expressing the MGMT gene prior to transplanting them into brain-tumor patients. This would allow for the patients to receive stronger doses of temozolomide, since the patient's hematopoietic cells would be resistant to the drug. High doses of temozolomide in high-grade gliomas have low toxicity, but the results are comparable to the standard doses.
Etacrynic acid (INN) or ethacrynic acid (USAN), trade name Edecrin, is a loop diuretic used to treat high blood pressure and the swelling caused by diseases like congestive heart failure, liver failure, and kidney failure. A disadvantage with ethacrynic acid, compared to the other loop diuretic drugs such as furosemide, is that it has a significantly steep dose-response curve. This means the drug's dosing is very important as small variance in dose can cause a significant difference in the biological response. Unlike the other loop diuretics, etacrynic acid is not a sulfonamide and may thus be used in individuals with sulfa allergies in whom the sulfonamide loop diuretics would be contraindicated.
=== Plants, climate and environmental change === Plant responses to climate and other environmental changes can inform one's understanding of how these changes affect ecosystem function and productivity. For example, plant phenology can be a useful proxy for temperature in historical climatology, and the biological impact of climate change and global warming. Palynology, the analysis of fossil pollen deposits in sediments from thousands or millions of years ago allows the reconstruction of past climates. Estimates of atmospheric CO2 concentrations since the Palaeozoic have been obtained from stomatal densities and the leaf shapes and sizes of ancient land plants. Ozone depletion can expose plants to higher levels of ultraviolet radiation-B (UV-B), resulting in lower growth rates. Moreover, information from studies of community ecology, plant systematics, and taxonomy is essential to understanding vegetation change, habitat destruction and species extinction.
Peppermint oil is under preliminary research for its potential as a short-term treatment for irritable bowel syndrome. High oral doses of peppermint oil (500 mg) can cause mucosal irritation and mimic heartburn. Peppermint oil capsules are licensed as a medicine in the UK for the treatment of irritable bowel syndrome (IBS). Preliminary research administering a tiny amount of the oil in order to lower the systolic aspect of blood pressure is in initial human trials in 2026. Peppermint oil and leaves have a cooling effect when used topically for muscle pain, nerve pain, relief from itching, or as a fragrance. Peppermint oil had supposed uses in ancient traditional medicine for minor gastrointestinal diseases.

The CUX1 gene Alternatively Spliced Product was first reported in 1997. The CUX1 gene has up to 33 exons. CASP mRNA includes exons 1 through 14 and 25 through 33. The human CASP protein is predicted to contain 678 amino acids, of which 400 are shared with CUTL1. CASP protein is approximately 80 kD. It lacks the DNA binding region of CUTL1, but instead contains a trans-membrane domain that allows it to insert into lipid bilayers. It has been localized to the Golgi apparatus. CASP has been reported to be part of a complex with Golgin 84 that tethers COPI vesicles and is important for retrograde transport in the Golgi and between the Golgi and endoplasmic reticulum. The targeting of vesicles involves tethers and SNAREs.
=== Later years === Macleod returned to Scotland in 1928 to become Regius Professor of Physiology at the University of Aberdeen (in succession to his former teacher, John Alexander MacWilliam who retired in 1927) and later Dean of the University of Aberdeen Medical Faculty. Between 1929 and 1933 he was also a member of the Medical Research Council. Macleod did not continue to work on insulin, but he remained active as a researcher, lecturer and author. His last major contribution was a proof that the central nervous system does have an important role in maintaining carbohydrate metabolism balance, as was his original hypothesis. His theory about conversion of fats into carbohydrates remained unproven, despite his provision of several indirect proofs. He devoted his spare time to golf, motorcycling and painting. He married Mary Watson McWalter (1876–1940) in 1903, but they never had children. He died in 1935 in Aberdeen after several years of suffering from arthritis, despite which he remained active almost until his death. In 1933 he made a lecture tour of the US, and in 1934 he published the 7th edition of his book Physiology and Biochemistry in Modern Medicine.
=== Imaging === Mammography is the method of choice for radiologic examination of male breast tissue in the diagnosis of gynecomastia when breast cancer is suspected on physical examination. If a mass/lump is felt during a physical exam, some features of the lump that would point to malignancy would be painless, non-movable (fixed), irregularly shaped, and skin changes. Mammography is rarely indicated for men since breast cancer is an unlikely diagnosis. If mammography is performed and does not reveal findings suggestive of breast cancer, further imaging is not typically necessary. If a tumor of the adrenal glands or the testes is thought to be responsible for the gynecomastia, ultrasound examination of these structures may be performed.

It is a synthetic peptide triagonist that engages GIP, GLP-1, and glucagon receptors. Investigational compounds in this class are studied for metabolic conditions rather than for a single organ system.
No regulatory agency has approved it for any indication. It remains an investigational product studied within controlled clinical trial programs.
A dual agonist engages two receptor targets, typically GIP and GLP-1. A triagonist adds glucagon receptor activity, which is intended to influence energy expenditure alongside appetite and glucose handling.
A responder analysis counts participants who cross a threshold, such as five or ten percent weight loss. It complements average percent change by showing how widely results are distributed. The two measures can diverge when a subset of participants loses a large amount.
