Quantification of the peptide in biological samples generally relies on liquid chromatography coupled with tandem mass spectrometry. This approach separates the analyte from matrix components and detects it by mass-to-charge transitions specific to the molecule. Immunoassays offer higher throughput but can cross-react with related peptides and metabolites, so mass spectrometric methods are preferred when structural confirmation is required. Method validation typically addresses accuracy, precision, selectivity, and stability under handling conditions.
Several questions remain unresolved. It is not yet known whether the compound reduces cardiovascular events or mortality, because outcome studies require long follow-up. The durability of weight reduction after treatment withdrawal is uncertain, and rebound has been observed with other incretin-based therapies. Long-term safety data covering several years are limited. Effects in adolescents, in pregnancy, and in people with significant kidney or liver impairment have not been characterized in published reports.
Randomized studies of retatrutide measure change in body weight as a percentage of baseline, along with absolute weight loss. Glycemic endpoints include hemoglobin A1c and fasting plasma glucose. Investigators also track blood pressure, lipid fractions, and liver fat content to characterize effects beyond weight alone. Trial designs typically use double-blind, placebo-controlled groups with periodic dose escalation, and they record adverse events throughout both treatment and follow-up periods.
Retatrutide is an investigational synthetic peptide that acts as an agonist at three distinct G protein-coupled receptors. It combines activity at the glucagon-like peptide-1 receptor, the glucose-dependent insulinotropic polypeptide receptor, and the glucagon receptor within a single molecule. This multi-receptor profile distinguishes it from earlier incretin-based compounds that engage one or two of these pathways. Researchers designed the molecule to test whether simultaneous activation produces greater metabolic effects than single or dual agonism alone.
The peptide backbone is chemically modified to resist rapid enzymatic breakdown in the body. A fatty acid side chain promotes binding to serum albumin, which slows renal clearance and supports an extended circulation time. These modifications allow less frequent administration than would be possible with an unmodified peptide. The precise contribution of glucagon receptor activation to the overall metabolic effect remains an area of active investigation, because glucagon raises glucose while also increasing energy expenditure.
Development has progressed through early- and mid-stage human studies in adults with obesity and with type 2 diabetes. Published phase 2 data reported reductions in body weight and improvements in glycemic markers over the treatment period. No regulatory agency has approved the compound for any indication, and it remains available only within controlled research settings. Whether benefits observed in trials translate into durable outcomes after treatment stops is not yet established.

Stability depends strongly on physical state. Dry powder is comparatively robust when held at -20 °C or below, desiccated and shielded from light; under those conditions degradation is slow and measured over years. Once dissolved, the peptide becomes far more vulnerable: backbone hydrolysis, oxidation of susceptible residues and aggregation all proceed faster in solution, and the rates climb with temperature and with pH far from neutral. Refrigerated storage at 2–8 °C extends usable life for short periods, and repeated freeze–thaw cycles are best avoided.
Identity and purity are established by instrumental methods rather than by appearance. Reversed-phase high-performance liquid chromatography separates the peptide from related impurities and yields a purity value, usually expressed as the share of total peak area. Mass spectrometry checks that the observed mass agrees with the mass calculated from the published sequence, while peptide mapping or amino acid analysis adds structural evidence. Water content, counter-ion identity and residual solvents are sometimes reported as well. A certificate of analysis should name the method behind each figure, because results are method-dependent.
Laboratory handling follows the conventions used for other synthetic peptides. Lyophilized material is weighed and dissolved in an aqueous diluent, typically sterile water or bacteriostatic water, using gentle swirling rather than vigorous shaking, because foaming stresses the chain. Solutions are prepared under clean conditions and, where sterility matters, passed through a suitable filter. Working portions are kept small so that stock material is not repeatedly warmed and cooled, a practice that limits both aggregation and gradual loss of activity.
Human evidence remains limited to controlled studies. A phase 2 trial in adults with obesity reported large, dose-dependent reductions in body weight over 48 weeks, with gastrointestinal events as the most frequently recorded adverse effect. Phase 3 programs designated TRIUMPH, for obesity, and TRANSCEND, for type 2 diabetes, are intended to confirm efficacy and to characterize safety in larger populations. Related studies are examining conditions such as knee osteoarthritis in people with obesity and metabolic liver disease. Open questions include long-term tolerability, effects on lean mass, and what happens after treatment is stopped.
Retatrutide is an investigational synthetic peptide that acts on three receptor targets at once: glucose-dependent insulinotropic polypeptide, glucagon-like peptide-1, and glucagon. It is developed by Eli Lilly and appears in the literature and in trial registries under the code LY3437943. The molecule belongs to a class of engineered peptides designed to resist rapid breakdown and permit infrequent subcutaneous administration. No regulatory agency has approved it for clinical use, and all available human data come from controlled trials rather than from routine practice.
The intended pharmacology combines three signals in one molecule. GLP-1 receptor activation reduces appetite and slows gastric emptying, effects already exploited by approved incretin-based therapies. GIP receptor engagement is associated with improved insulin sensitivity and with direct effects on adipose tissue, although how much it contributes to overall outcomes is still debated. Glucagon receptor agonism raises energy expenditure and supports hepatic lipid handling, a mechanism that also tends to increase glucose output. The triple profile is hypothesized to produce a larger metabolic effect than single or dual agonism, but the relative weight of each receptor in humans is not settled.
As a peptide, the compound is generally supplied as a lyophilized powder and stored frozen to slow degradation. Recommended conditions usually sit at minus twenty degrees Celsius or colder, shielded from light and moisture. Solutions are less stable than the dry powder and are often prepared fresh before analysis. Repeated freeze-thaw cycles can drive aggregation, so splitting stock material into small aliquots reduces handling stress and preserves sample integrity.
Quality control of research material relies on several complementary checks. Purity testing confirms the absence of truncated or oxidized peptide species, while water content and counterion analysis show how much mass comes from salts rather than the peptide itself. Sequence verification through tandem mass spectrometry ensures the correct amino acid chain. Because unregulated suppliers vary widely, independent verification of identity and purity is often necessary before a sample enters experiments.
III (stylized as (III)) is the third studio album by Canadian electronic music duo Crystal Castles, released on November 7, 2012, by Fiction Records and Polydor Records. Production was handled by Ethan Kath, with additional production by Jacknife Lee. III is the last Crystal Castles album with vocalist Alice Glass before her departure in 2014, and is also their last eponymous album, as the band's 2016 album is titled Amnesty (I) instead of (IV) or Crystal Castles. Glass continued the series on her own by including "IV" into the title of her 2022 debut album Prey//IV.
=== Magazine === OECD Observer, an award-winning magazine, was launched in 1962. The magazine appeared six times a year until 2010, and became quarterly in 2011 with the introduction of the OECD Yearbook, launched for the 50th anniversary of the organisation. The online and mobile editions contained news, analysis, reviews, commentaries and data on global economic, social and environmental challenges and listings of the latest OECD books. An OECD Observer Crossword was introduced in Q2 2013. The OECD Observer was last issued in the fourth quarter of 2019, with a double edition looking ahead at artificial intelligence, and a cover leading on why statistical offices should hire a comedian. The OECD Observer website closed in the first quarter of 2021; the archive can be consulted at www.oecd.org.
N-Acetylcysteine ethyl ester (NACET, NAC-Et) is an antioxidant which acts as a lipophilic prodrug for N-acetylcysteine and subsequently glutathione, with improved bioavailability over N-acetylcysteine itself, and has been researched in animal studies as a potential treatment for diabetic retinopathy. It has also been sold as a "dietary supplement" in combination with glycine under the name GlyNAC-Et as a supposedly improved version of the older glycine-NAC combination GlyNAC which has been claimed to show efficacy in counteracting symptoms associated with aging as well as HIV infection, diabetes and nerve damage. Several related compounds such as γ-glutamylcysteine ethyl ester and glutathione ethyl ester have been investigated for similar applications.

Members of the group held no voting privileges, so they could not affect the outcome of the parent group. The Order evolved into other fraternal groups during the 1920s. Teepee Order of America - Founded in 1915 by Red Fox Francis St. James, an alleged Blackfoot Indian. Open to Indians and non-Indians from the US, Canada, and Latin America, it excluded blacks and European immigrants. Originally similar to the Boy Scouts of America in appealing to youth and "focusing on Indian activities and ceremonies that were romantic in nature." It was a Pan-Indian group, but not as successful as the Society of American Indians, though there was some membership overlap. It advocated for Native American citizenship and questioned the efficacy of the Bureau of Indian Affairs.
In reconstructive rhinoplasty, the defects and deformities that the plastic surgeon encounters, and must restore to normal function, form, and appearance include broken and displaced nasal bones; disrupted and displaced nasal cartilages; a collapsed bridge of the nose; congenital defect, trauma (blunt, penetrating, blast), autoimmune disorder, cancer, intranasal drug-abuse damages, and failed primary rhinoplasty outcomes. Rhinoplasty reduces bony humps, and re-aligns the nasal bones after they are cut (dissected, resected). When cartilage is disrupted, suturing for re-suspension (structural support), or the use of cartilage grafts to camouflage a depression allow the re-establishment of the normal nasal contour of the nose for the patient. When the bridge of the nose is collapsed, rib-cartilage, ear-cartilage, or cranial-bone grafts can be used to restore its anatomic integrity, and thus the aesthetic continuity of the nose. For augmenting the nasal dorsum, autologous cartilage and bone grafts are preferred to (artificial) nose prostheses, because of the reduced incidence of histologic rejection and medical complications.
=== Metabolic actions === Several SIRT2 deacetylation targets play important roles in metabolic homeostasis. SIRT2 inhibits adipogenesis by deacetylating FOXO1 and thus may protect against insulin resistance. SIRT2 sensitizes cells to the action of insulin by physically interacting with and activating Akt and downstream targets. SIRT2 mediates mitochondrial biogenesis by deacetylating PGC-1α, upregulates antioxidant enzyme expression by deacetylating FOXO3a, and thereby reduces ROS levels. Also, Sirt2 can reactivate the inactive G6PD by removing the acetyaltion at K403.
The authoritarian Sanation government ("sanation" meant to denote "healing") that Piłsudski led until his death in 1935 (and would remain in place until 1939) reflected the dictator's evolution from his center-left past to conservative alliances. Political institutions and parties were allowed to function, but the electoral process was manipulated and those not willing to cooperate submissively were subjected to repression. From 1930, persistent opponents of the government, many of the leftist persuasion, were imprisoned and subjected to staged legal processes with harsh sentences, such as the Brest trials, or else detained in the Bereza Kartuska prison and similar camps for political prisoners. About three thousand were detained without trial at different times at the Bereza internment camp between 1934 and 1939. In 1936 for example, 369 activists were taken there, including 342 Polish communists. Rebellious peasants staged riots in 1932, 1933 and the 1937 peasant strike in Poland. Other civil disturbances were caused by striking industrial workers (e.g. events of the "Bloody Spring" of 1936), nationalist Ukrainians[p] and the activists of the incipient Belarusian movement. All became targets of ruthless police-military pacification.[y] Besides sponsoring political repression, the government fostered Józef Piłsudski's cult of personality that had already existed long before he assumed dictatorial powers.

As such, the transdermal route cannot easily achieve adequate circulating progesterone levels, and this makes transdermal progesterone impractical for systemic therapy. Clinical studies have found only very low circulating levels of progesterone with the use of transdermal progesterone, and these levels are thought to be insufficient to confer endometrial protection against estrogens. The range of circulating levels of progesterone that has been observed in clinical studies with various formulations and doses of transdermal progesterone is 0.38 to 3.5 ng/mL. Although very low levels of progesterone have been observed in venous blood with transdermal progesterone, very high and in fact greatly supraphysiological levels of progesterone have unexpectedly been found in saliva and capillary blood. In one study, the levels of progesterone in saliva and capillary blood were 10- and 100-fold greater than levels in venous blood, respectively. Levels of salivary progesterone that have been observed have ranged from 2.9 to 2,840 ng/mL. The high salivary and capillary blood levels of progesterone suggest that despite low circulating levels of progesterone, systemic distribution of progesterone and considerable exposure of some tissues to the hormone may be occurring with transdermal progesterone somehow. However, the few clinical studies that have assessed the effects of transdermal progesterone on the endometrium have had mixed findings, and further research is needed to determine whether it can confer adequate endometrial protection as a component of menopausal hormone therapy.
=== 13 July === Ten people were killed in SAF airstrikes in Abu Zabad and Al-Fulah in West Kordofan. The RSF claimed to have taken the town of Um Sumaima in North Darfur. However, Darfur's governor, Minni Minnawi, claimed that it had been retaken by the JDF. The bodies of 26 people killed by the RSF were recovered from a mass grave at the Al-Nashisheiba campus of the University of Gezira.
Ortho Clinical Diagnostics (now QuidelOrtho) is an in vitro diagnostics company that made products and diagnostic equipment for blood testing. Ortho served two primary industries in the medical field: clinical laboratories, by producing platforms and assays that test for a variety of diseases, conditions, and substances; and immunohematology, by providing the means to ensure blood transfusion recipients receive appropriate and compatible blood. Johnson and Johnson acquired Eastman Kodak's Clinical Diagnostics Division in 1994 (to form Johnson & Johnson Clinical Diagnostics), which was then merged with Ortho Diagnostic Systems in 1997. QuidelOrtho's global corporate offices are in Raritan, New Jersey, while their global research and development center is in Rochester, New York. In 2014, The Carlyle Group purchased the company from Johnson & Johnson for $4.15 billion. Ortho Clinical Diagnostics now operated as an independent company, up until its acquisition by Quidel Corporation for $6 billion, on May 27, 2022.

University of Kentucky plastic surgeon Brian Rinker encountered many women in his practice who attributed their sagging breasts to breastfeeding, which was also the usual belief among medical practitioners. He decided to find out if this was true, and between 1998 and 2006 he and other researchers interviewed 132 women who were seeking breast augmentation or breast lifts. They studied the women's medical history, body mass index (BMI), their number of pregnancies, their breast cup size before pregnancy, and smoking status. The study results were presented at a conference of the American Society of Plastic Surgeons. According to Rinker's research, there are several key factors. A history of cigarette smoking "breaks down a protein in the skin called elastin, which gives youthful skin its elastic appearance and supports the breast." The number of pregnancies was strongly correlated with ptosis, with the effects increasing with each pregnancy. As most women age, breasts naturally yield to gravity and tend to sag and fold over the inframammary crease, the lower attachment point to the chest wall. This is more true for larger-breasted women. The fourth reason was significant weight gain or loss (greater than 50 pounds (23 kg)). Other significant factors were higher body mass index and larger bra cup size. In Rinker's study, 55% of respondents reported an adverse change in breast shape after pregnancy. Many women mistakenly attribute the changes and their sagging breasts to breastfeeding, and as a result some are reluctant to nurse their infants.

Trials measure percentage change in body weight, absolute weight loss, and glycemic markers such as hemoglobin A1c. They also record blood pressure, lipids, and liver fat. Adverse events are tracked throughout.
Mass spectrometry identifies molecules by mass-to-charge transitions, which reduces interference from related peptides. Immunoassays run faster but can cross-react. Structural confirmation usually requires the mass spectrometric approach.
Cardiovascular outcomes, long-term safety, and weight regain after stopping treatment are unresolved. These questions need years of follow-up data. Published evidence covers only limited treatment durations.
